Abstract

The analysis of DNA is an essential tool in a variety of fields ranging from forensic sciences, medical research, to biological sciences. DNA karyotype is one of the tools used in recent years to' analyze DNA. DNA karyotype technique has been used to isolate and characterize organisms' genomic size. The purpose of this experiment was to isolate DNA from Aspergillus sp. and Penicillium sp., compared and characterized its genomic size by using pulse field gel electrophoresis (PFGE). To successfully karotype fungi DNA, the following procedures were done: (i) Prepared potato dextrose agar and potato dextrose broth for fungi growth, (ii) Monitored fungi growth, (iii) Prepared agarose gel for PFGE, and (iv) Prepared fungi chromosomes for PFGE. At the end of this research, the different chromosome sizes of Penicillium sp. and Aspergilus sp. was separated, viewed and characterized accordingly.
